-44-MI Paste Plus1261 In Vitro Root Surface Caries: Agents With and Without XylitolPoster Presentation3:45 PM–5:00 PM Mar 23, 2017CC, First Floor Authors:Authors:Catherine Flaitz(Presenter)Nationwide Children's Hospital and The Ohio State UniversityFranklin Garcia-Godoy, University of TennesseeJohn Hicks, Baylor College of MedicineAbstract: Objectives: Effects of xylitoldentifrice with sodium fluoride (Xylitol-NaF) on in vitro root caries formation were compared with commercially available NaFdentifrice and Casein Phosphopeptide-formation were compared with commercially available NaFdentifrice and Casein Phosphopeptide-Amorphous Calcium Phosphate with NaF(CPP-ACP) paste.Methods: Root surfaces from caries-free permanent teeth (n=10) underwent debridement and a fluoride-free prophylaxis. The tooth roots were sectioned into quarters, and acid-resistant varnish was placed with 2 sound root surface windows exposed on each tooth quarter. Each quarter from a single tooth was assigned to a treatment group: 1) No Treatment Control (synthetic saliva treatment only); 2) NaFDentifrice (Crest Cavity Protection Regular Paste, 0.243% Sodium Fluoride [NaF], Procter & Gamble); 3) Xylitol-NaFDentifrice (MyntSmile, 0.2% xylitol, 0.243% NaF, MyntSmileOral Health Care Products); and 4) CPP-ACP-FL (MI Paste Plus, xylitol, 0.243% NaF, MyntSmileOral Health Care Products); and 4) CPP-ACP-FL (MI Paste Plus, 0.2% NaF, CPP-ACP 10%, GC America). Tooth root quarters were treated with dentifrice twice daily (180s) followed by synthetic saliva rinsing over 7d period. In vitroroot caries were created using acidified gel (pH 4.25, 21 days). Longitudinal sections (3/tooth quarter, 60/group) were evaluated for mean lesion depths (water imbibition, polarized light, ANOVA, DMR).Results: Mean lesion depths were 332 ±29 µm No Treatment Control; 258 ±23 µm NaFDentifrice; 247 ±33 µm Xylitol-NaFdentifrice; 178 ±21 µm CPP-ACP-FL. All treatment agents had mean lesion depths significantly less than No Treatment Controls (P<.05). There was no significant difference in mean lesion depths between Xylitol-NaFand NaFdentifrices (P>.05). significant difference in mean lesion depths between Xylitol-NaFand NaFdentifrices (P>.05). However, CPP-ACP-FL provided a significant decrease in mean lesion depth compared with the other agents (P<.05)Conclusions: Fluoridated dentifrices provided significant reductions in root caries lesion depths compared with root surfaces not exposed to dentifrice treatment (P<0.05). Xylitol-NaFdentifrice reduced lesion depth similar to a commercially available NaFdentifrice (P>0.05). CPP-ACP-Fl had the greatest effect on caries reduction among treatment agents (P<.05). However, the in vitro physicochemical caries system employed could not assess additional beneficial effects of xylitolon biofilmsand reduction in acid production by cariogenicbacteria. CPP-ACP-Fl had greatest on biofilmsand reduction in acid production by cariogenicbacteria.
